flot1抑制胼胝质沉积促进南方根结线虫巨细胞发育

摘要:巨细胞是根结线虫定植植物根部,吸取宿主养分的关键结构。膜筏在细胞信号传导、细胞骨架稳定性和物质转运等过程中起着重要作用,可能与巨细胞形成及发育关系密切,但目前缺少膜筏与巨细胞相关研究。本文利用品红染色、戊二醛染色及定量pcr等技术,探究flot1缺失突变体及flot1-oe过表达拟南芥植株线虫侵染病理、生理表型,结果显示线虫侵染后flot1表达量升高,flot1缺失会显著降低拟南芥中线虫侵染数目,延缓线虫的生长发育,flot1缺失后较野生型拟南芥其巨细胞体积扩展受到明显抑制,根结中胼胝质合成基因表达上调,胼胝质含量显著升高且根部蔗糖含量显著降低。该结果表明,根结线虫可能诱导flot1表达上调,抑制胼胝质沉积并促进巨细胞发育。

flot1 inhibits callose deposition and promotes the development of giant cells in meloidogyne incognita
abstract:giant cells are key structures in the establishment of meloidogyne incognita in plant roots, playing a crucial role in absorbing nutrients from the host. lipid rafts play an important role in processes such as cell signaling, cytoskeletal stability, and material transport, and may be closely related to the formation and development of giant cells. however, research on the relationship between lipid rafts and giant cells is currently lacking. this study used techniques such as fuchsine staining, glutaraldehyde staining, and quantitative pcr to explore the pathological and physiological phenotypes of root-knot nematode infection in flot1 knockout mutants and flot1-oe overexpressing arabidopsis plants. the results showed that flot1 expression increased after nematode infection. the absence of flot1 significantly reduced the number of nematodes infecting arabidopsis and delayed nematode growth and development. after flot1 deletion, the expansion of giant cell volume in arabidopsis was significantly inhibited compared to the wild-type, the expression of callose synthesis genes in the root knot was upregulated, callose content significantly increased, and the sucrose content in the roots was notably reduced. these results suggest that meloidogyne incognita may induce the upregulation of flot1 expression, inhibit callose deposition, and promote the development of giant cells
