间歇性禁食改善类风湿性关节炎

摘要:目的:旨在探究间歇性禁食对关节滑膜成纤维细胞的影响及其在改善类风湿性关节炎中的作用。方法:通过 0 天、21 天对 dba/1j 小鼠尾根部皮内注射由胶原和佐剂等体积乳化的乳剂,以诱导小鼠自发性关节炎,同窝小鼠作为健康对照。运用关节炎评分评估关节炎小鼠的发病程度;通过western blotting和rt-qpcr检测小鼠肿胀后肢的炎症因子和基质金属蛋白酶(matrix metalloproteinases, mmps)表达水平;通过聚焦关节滑膜成纤维细胞的转录组差异基因,探究禁食对关节炎的改善作用通路。结果:禁食减缓关节炎小鼠的骨基质降解进展,并降低滑膜成纤维细胞对白细胞的募集作用,从而改善关节骨损伤。

abstract:objective: to explore the effects of intermittent fasting on synovial fibroblasts and its role in improving rheumatoid arthritis. methods: dba/1j mice were injected intradermally at the base of the tail with an emulsion of equal volumes of collagen and adjuvant on day 0 and day 21 to induce spontaneous arthritis in mice, with littermates serving as healthy controls. arthritis severity was evaluated using arthritis scores; the expression levels of inflammatory factors and matrix metalloproteinases in the swollen hind limbs of mice were detected by western blotting and rt-qpcr; and the pathways by which fasting improves arthritis were explored by focusing on the transcriptional differences in synovial fibroblasts. results: intermittent fasting slowed the progression of bone matrix degradation in arthritic mice and reduced the recruitment of leukocytes by synovial fibroblasts, thereby improving joint bone damage.
