凝血因子xii的二硫键在蛋白合成、分泌与功能的研究

摘要:凝血因子xii是内源性凝血途径的起点,通过激活fxi介导凝血酶的生成。尽管研究指出它在止血过程中的作用相对微弱,但凝血因子xii在病理性血栓形成中却扮演着举足轻重的角色。正因如此,它已成为当前抗血栓治疗药物研发中的一个重要且安全的靶点。凝血因子xii的结构已经得到了完整的解析,但是20个二硫键的功能缺乏研究,目前仅有少量病例报告涉及,因此本文对凝血因子xii的二硫键在蛋白合成、分泌与功能中的作用进行完整的解析,从而为基于其结构设计的新型抗凝药物奠定坚实的理论基础。

characterization on disulfide bond of coagulation factor xii for protein synthesis, secretion and function
abstract:coagulation factor xii is the starting point of the endogenous clotting pathway and mediates the production of thrombin by activating fxi. it has been found that coagulation factor xii plays a weak role in hemostasis, but it plays an important role in pathological thrombosis. therefore, coagulation factor xii is a safe target site for antithrombotic therapeutics at present. the structure of coagulation factor xii has been fully analyzed, but the function of disulfide bonds has been poorly studied, and only some cases have been reported. therefore, the role of disulfide bonds of coagulation factor xii in protein synthesis, secretion and function have been completely analyzed in this paper, providing a basis for the design of anticoagulant drugs based on the structure of coagulation factor xii.
