lmo2调控etp-all细胞生长的研究

摘要:目的:早期t细胞前体急性淋巴白血病(early t-cell precursor acute lymphoblastic leukemia,etp-all)是近年才被鉴定为t-all新亚型,表现为lmo2、lyl1和hhex高表达。其中,lmo2(lim-onlyprotein2)作为lyl1、hhex的转录因子对于etp-all疾病的发生发展具有重要作用。本研究将检测lmo2在etp-all细胞中的表达,并研究它对etp-all细胞生长的影响。方法:(1)从geo数据库中下载人类与小鼠的淋巴祖细胞rna-seq数据集,通过netbid分析lmo2的调控活性;(2)通过检索cancer cell line encyclopedia(ccle)数据库分析多种癌症细胞系中lmo2的表达,同时蛋白质免疫印迹(western blotting,wb)检测etp-all细胞系与t-all细胞系中lmo2的表达;(3)设计针对人源lmo2的shrna,利用慢病毒侵染方式,在loucy细胞中稳定敲低lmo2表达,检测其对loucy细胞体外生长、集落生成能力和etp-all相关基因表达水平的影响;结果:(1)lmo2在t细胞发育过程中表达逐渐降低;(2)相比于其他癌症细胞系,lmo2在etp-all细胞系中呈现高表达;(3)敲减lmo2抑制etp-all细胞的细胞增殖和集落生成能力,并促进凋亡,同时降低etp-all相关基因的表达水平;结论:lmo2在etp-all中高表达并调控etp-all细胞的生长。

lmo2 regulates the growth of etp-all cells
abstract:objective: early t-cell precursor acute lymphoblastic leukemia (etp-all) is a new subtype of t-all identified only in recent years, which exhibits high expression of lmo2, lyl1 and hhex. among them, lmo2 (lim-onlyprotein2), as a transcription factor of lyl1 and hhex, plays an important role in the development of etp-all disease. in this study, we will detect the expression of lmo2 in etp-all cells and investigate its effect on the growth of etp-all cells. methods: (1) lymphoid progenitor cell rna-seq datasets oflmo2 regulates the growth of etp-all cells human and mouse were downloaded from the geo database, and the regulatory activity of lmo2 were analyzed by using netbid; (2) the expression of lmo2 in various cancer cell lines was analyzed through querying the cancer cell line encyclopedia (ccle) database, and meanwhile the expression of lmo2 in etp-all cell lines and t-all cell lines were detected by using western blotting (wb); (3) two independent short-hairpin rnas(shrnas) against lmo2 and the control were delivered into loucy cells with lentiviral vectors. the expression of lmo2 in these cells was analyzed by western blotting. the effect of lmo2 silencing on the growth, the colony-forming cell(cfc) production and the expression pf etp-all- related genes were studied; results: (1) the expression of lmo2 gradually decreased during t cell development; (2) compared to other cancer cell lines, lmo2 showed high expression in etp-all cell line. (3) lmo2 silencing significantly inhibited the growth, colony formation and expression of etp-all-related genes in loucy cells; conclusion: lmo2 is highly expressed in etp-all and regulates the growth of etp-all cells.
