摘要:目的:研究lncrna neat1在shh型髓母细胞瘤发生发展中所起的作用及调控shh信号通路可能的机制,为shh型髓母细胞瘤的治疗探索新的靶点。方法:使用分子生物学技术检测neat1在shh型髓母细胞瘤中所起的作用、对shh信号通路的调控作用以及具体的调控机制。结果:1)neat1在shh型髓母细胞瘤中高表达;2)neat1可以调控shh型髓母细胞瘤的增殖;3)neat1可以调控shh信号通路;4)neat1通过mir-344-5p/gli1轴影响shh型髓母细胞瘤的发展。结论:neat1能够特异性地抑制mir-344-5p的表达,从而间接抑制了mir-344-5p对gli1 mrna的转录后调控,使得gli1蛋白表达增高,揭示了neat1/mir-344-5p/gli1调控通路在shh型髓母细胞瘤发生发展中的作用机制。

neat1/mir-344-5p/gli1 promotes the progression of shh medulloblastoma
abstract: objective: to investigate the role of lncrna neat1 in the occurrence and development of shh medulloblastoma and the possible mechanism of regulating shh signaling pathway, and to explore new targets for the treatment of shh medulloblastoma. methods: molecular biology experiments were used to detect the role of neat1 in shh medulloblastoma, its role in shh signaling pathway, and the specific regulatory mechanism. results: 1) neat1 was highly expressed in shh medulloblastoma; 2) neat1 can regulate the proliferation of shh medulloblastoma; 3) neat1 can regulate shh signaling pathway; 4) neat1 affects the development of shh medulloblastoma through the mir-344-5p/gli1 axis. conclusion: neat1 can specifically inhibit the expression of mir-344-5p, thereby indirectly inhibiting the post-transcriptional regulation of gli1 mrna by mir-344-5p, increasing the expression of gli1 protein.the study reveal the mechanism of neat1/mir-344-5p/gli1 regulatory pathway in the occurrence and development of shh medulloblastoma.
