滨蒿内酯通过调节egfr/mapk通路诱导卵巢癌细胞凋亡

摘要:目的:探究滨蒿内酯对卵巢癌细胞功能的影响。方法:分别通过mtt、克隆集落检测滨蒿内酯对卵巢癌细胞系增殖能力的影响,利用划痕、transwell侵袭实验检测滨蒿内酯对卵巢癌细胞系迁移侵袭能力的影响,通过wb、流式细胞术评估滨蒿内酯处理后卵巢癌细胞的凋亡水平,利用在线网站swiss预测滨蒿内酯潜在结合蛋白,通过go和kegg分析富集蛋白通路,利用wb技术检验滨蒿内酯结合蛋白预测结果。结果:mtt和克隆集落结果显示滨蒿内酯有效抑制卵巢癌细胞的增殖能力,划痕和侵袭实验结果表明滨蒿内酯显著抑制卵巢癌的迁移和侵袭能力,wb和流式细胞结果显示,滨蒿内酯能够有效促进卵巢癌细胞的凋亡水平,并且swiss在线分析网站显示滨蒿内酯潜在结合蛋白与凋亡信号通路重叠,经过wb检验,滨蒿内酯调控egfr/mapk信号通路。结论:滨蒿内酯显著抑制卵巢癌细胞的增殖、迁移和侵袭水平,并且通过egfr/mapk信号通路促进卵巢癌细胞的凋亡。

scoparone induces ovarian cancer cell apoptosis induces apoptosis
abstract:objective: to investigate the effects of scoparone on the functions of ovarian cancer cells. methods: mtt and colony formation assays were used to examine the effects of scoparone on the proliferation ability of ovarian cancer cell lines. scratch wound healing assay and transwell invasion assay were performed to detect the effects of scoparone on the migration and invasion abilities of ovarian cancer cell lines. western blot (wb) and flow cytometry were employed to evaluate the apoptosis level of ovarian cancer cells after scoparone treatment. the swiss online website was used to predict the potential binding proteins of scoparone, and gene ontology (go) and kyoto encyclopedia of genes and genomes (kegg) analyses were conducted to enrich protein pathways. the predicted results of scoparone-binding proteins were verified by wb technology. results: mtt and colony formation assays revealed that scoparone effectively inhibited the proliferation ability of ovarian cancer cells. scratch wound healing and invasion assays showed that scoparone significantly suppressed the migration and invasion abilities of ovarian cancer. wb and flow cytometry results indicated that scoparone effectively promoted the apoptosis level of ovarian cancer cells. the swiss online analysis website revealed that the potential binding proteins of scoparone overlapped with the apoptosis signaling pathway. after wb verification, scoparone regulated the egfr/mapk signaling pathway. conclusion: scoparone significantly inhibits the proliferation, migration, and invasion levels of ovarian cancer cells and promotes apoptosis through the egfr/mapk signaling pathway.
