maff在骨骼肌缺血再灌注损伤修复中的作用及其潜在机制

摘要:目的:探讨maff(maf bzip transcription factor f, maff)参与骨骼肌再生的作用方式及相关机制。方法:利用野生型卫星细胞以及构建缺血再灌注(ischemia reperfusion, ir)损伤模型,探究maff的细胞定位以及在ir损伤后表达变化情况;构建过表达maff的c2c12成肌细胞系,探究maff对c2c12成肌细胞增殖和分化的影响;利用chip、双荧光素酶报告实验,探究研究maff与肌源性相关因子的调控关系。结果:maff主要表达于卫星细胞核中,并在ir损伤后瞬时上调,提示maff参与骨骼肌ir损伤修复过程;过表达maff后不影响c2c12成肌细胞的增殖,且chip实验显示maff不能靶向调控pax7(paired box protein 7, pax7)表达,提示maff参与肌肉损伤再生机制与c2c12成肌细胞的增殖无关;myod(myogenic determining factor, myod),可结合于maff启动子;过表达maff后发现maff与myogenin(myogenin, myog)具有正相关性,并可靶向结合并激活myogenin的表达。结论:maff可通过参与myod/maff/myogenin通路促进卫星细胞的分化,对肌肉再生能力起到正调控的作用。

the role and mechanism of maff in the repair of skeletal muscle ischemia reperfusion injury
abstract:objective: the aim of this study is to investigate the role and its underlying mechanism of maff in muscle regeneration. methods: by using wild-type satellite cells and constructing ischemia-reperfusion injury model, the localization of maff cells and the change of its expression after ir injury were investigated. the effect of maff on proliferation and differentiation of c2c12 myoblasts was investigated by constructing a maff overexpressing c2c12 myoblast line. through chip and dual luciferase reporting experiments, the regulatory relationship between maff and myogenic related factors was investigated. results: maff is expressed in satellite nucleus and is up-regulated immediately after ir injury, suggesting that maff is involved in the repair process of ir injury in skeletal muscle. overexpression of maff did not affect the proliferation of c2c12 cells, and chip experiments showed that maff could not target the expression of pax7, suggesting that maff\'s involvement in muscle injury and regeneration is not associated with the proliferation of satellite cells. myod, a determinant of early differentiation of satellite cells, can bind to the maff promoter. after overexpression of maff, it was found that maff was positively correlated with the late differentiation factor myogenin, and could target and activate the expression of myogenin.
